Novel mutation c.1210-3C>G in with a poly-T tract of 5T affects mRNA splicing in a Chinese patient

Frontiers of Medicine 2022, Volume 16, Issue 1,   Pages 150-155 doi: 10.1007/s11684-021-0846-5

Abstract: Cystic fibrosis (CF) is a rare autosomal recessive disease with only one pathogenic gene cystic fibrosis transmembrane conductance regulator (CFTR). To identify the potential pathogenic mutations in a Chinese patient with CF, we conducted Sanger sequencing on the genomic DNA of the patient and his parents and detected all 27 coding exons of CFTR and their flanking intronic regions. The patient is a compound heterozygote of c.2909G>A, p.Gly970Asp in exon 18 and c.1210-3C>G in cis with a poly-T of 5T (T5) sequence, 3 bp upstream in intron 9. The splicing effect of c.1210-3C>G was verified via minigene assay in vitro, indicating that wild-type plasmid containing c.1210-3C together with T7 sequence produced a normal transcript and partial exon 10-skipping-transcript, whereas mutant plasmid containing c.1210-3G in cis with T5 sequence caused almost all mRNA to skip exon 10. Overall, c.1210-3C>G, the newly identified pathogenic mutation in our patient, in combination with T5 sequence in cis, affects the CFTR gene splicing and produces nearly no normal transcript in vitro. Moreover, this patient carries a p.Gly970Asp mutation, thus confirming the high-frequency of this mutation in Chinese patients with CF.

Jointly optimized congestion control, forwarding strategy, and link scheduling in a named-data multihop

Cheng-cheng Li, Ren-chao Xie, Tao Huang, Yun-jie Liu,lengcangche@bupt.edu.cn,renchao_xie@bupt.edu.cn,htao@bupt.edu.cn

Frontiers of Information Technology & Electronic Engineering 2017, Volume 18, Issue 10,   Pages 1573-1590 doi: 10.1631/FITEE.1601585

Abstract: As a promising future network architecture, named data networking (NDN) has been widely considered as a very appropriate network protocol for the (MWN). In named-data MWNs, is a critical issue. Independent optimization for may cause severe performance degradation if it can not cooperate well with protocols in other layers. Cross-layer is a potential method to enhance performance. There have been many cross-layer mechanisms for MWN with Internet Protocol (IP). However, these cross-layer mechanisms for MWNs with IP are not applicable to named-data MWNs because the communication characteristics of NDN are different from those of IP. In this paper, we study the joint , forwarding strategy, and link scheduling problem for named-data MWNs. The problem is modeled as a network utility maximization (NUM) problem. Based on the approximate subgradient algorithm, we propose an algorithm called ‘jointly optimized , forwarding strategy, and link scheduling (JOCFS)’ to solve the NUM problem distributively and iteratively. To the best of our knowledge, our proposal is the first cross-layer mechanism for named-data MWNs. By comparison with the existing mechanism, JOCFS can achieve a better performance in terms of network throughput, fairness, and the pending interest table (PIT) size.

New Mode of City Without Road Congestion—Saving 70% of Land, Capital and Energy

Dong Guoliang

Strategic Study of CAE 2006, Volume 8, Issue 1,   Pages 11-17

Abstract:

This paper puts forward “Ideas on Reforming City Gene” and “New Smooth City Mode”. At present, city suffers from fatal gene —“misplaced space”,which leads to ten serious problems hindering city sustainable development: occupation of too much city land, low efficiency, high energy consumption, heavy pollution, great difficulty in parking, poor public security, uncomfortable living environment, frequent occurrence of traffic accidents and huge amount of transportation investment. It is better to carry out study from perspective of 4 levels of “city transportation—city space—economy—society”,locate transportation problems from other point of view instead of from transportation, and get rid of gene of “misplaced space”. Only by this way, can many problems concerning city transportation and sustainable development get solved completely. A disordered city space can be separated into two free spaces by constructing one for cars and one for people respectively, thus constructing a “New Smooth City Mode”. The new mode can realize goal of finding new street system to adapt to characteristics of car transportation as put forward by “Athens Charter” as well as target of building saving and pleasant “human-oriented” city.

Real-time dynamic evaluation of urban netword

Quan Yongshen,Guo Jifu,Wen Huimin,Sun Jianping

Strategic Study of CAE 2011, Volume 13, Issue 1,   Pages 43-48

Abstract:

This paper analyzes the dynamic nature of real-time traffic network performance measurement, with particular focus on random traffic flow fluctuation, traffic deferred conduction effect and traffic periodicity. The limitations of the traditional traffic network evaluation methods based on traffic V/C is discussed; followed by the presentation of a novel approach for real-time traffic network performance evaluation, which solves the previous technical issues such as data collection and processing with no blind region, dynamic spatial-temporal traffic network performance analysis and evaluation threshold calibration. The proposed approach is validated in a case study using the Beijing traffic network, which proves its rationality, efficiency and practicality. The implications of the research and methodologies presented in this paper provide new theories and perspectives for infrastructural planning, dynamic road network performance evaluation, etc.
